Micropropagation

Micropropagation is the cultivation of small plant segments or explants in liquid or gel medium for the purpose of clonal multiplication of plant material. Some plants are difficult to propagate vegetatively by cuttings and sometimes micropropagation is suitable for these species. This outlines a project for the micropropagation of hazelnut. Further a temporary immersion system is detailed.
Plants Were Grown Indoors Under Artificial Light to Minimize Contaminating Fungi and Bacteria


Small Segments Containing One Node Were Cut And Sanitized
    Sanitization involved washing plant material with 70% ethanol for 30 seconds followed by 10 minutes of a 10% bleach wash with one drop of detergent. The plant segments were agitated in the bleach solution over the course of the wash period. The plant segments are then triple-rinsed in sterile autoclaved water. Apical buds were not sucessful, so only nodes with axillary buds are used here. Segments are then trimmed to freshen the cut end and then inserted into a DKW nutrient agar and incubated at 75 deg with a 16h/8h light dark cycle under LED shop lights.

Segments were then placed in new test tubes for regrowth or placed in bulk into a temporary liquid immersion system.

  • 6-Benzylaminopurine - Growth Hormone

  • Plant Preservation Mixture - antimicrobial 












  • Xacto Knife or Scalpel





  • Ascorbic Acid (vitamin C powder) - prevents browning of tissues




  • Gibberellic Acid - Growth Hormone




  • Hefty Brand Container with tight fitting lid - for clean box aseptic working area




  • Seed Starting Mat and Thermostat - For temperature control




  • Sterilite 32 quart gasketed lid box - for incubating test tubes

  • Ethanol - sterilizing tools


Bleach - sterilizing plant material


Mason Jars - sterilizing tools and plant material


Scale with accuracy to .01 g - weighing ingredients


1ml Syringe - measuring liquid ingredients


Agar Powder - gelling agent for medium


4 oz Amber Bottles - diluting and storing hormones


Pressure Cooker - Sterilizing Medium and Rinse Water


  Outlet Timer- light timing control
